For example, for a fundies hedge fund or private equity, typically you work at an investment bank before moving over to a hedge fund or private equity firm. Is this true of being a quant also - i.e., you would need to work in some sort of sell-side role before moving over to a quant hedge fund or prop firm?

Yes, you can, I would bet Ph.D. in hard science is much better than MFE.
Almost every big name quant fund hires from Ph.D.
